Stratégie de marquage covalent du récepteur P2X7 natif pour microscopie super- résolutive

Abstract : P2X7, a ligand-gated ion channel activated by extracellular ATP, is a major feature of purinergic signaling. Its functions is yet to be elucidated, notably in the central nervous system homeostasis balance. Its neuropathological implications are numerous and highly related to inflammation and its propagation. We are currently lacking selective tools to decipher the fine neuropathological dynamic on this protein. The current work here presents the development and application of an innovative chemical approach base on affinity to covalently decorate this receptor with a biotin. The final reagent, called LNASAB, allows us to perform super-resolution in microglia in different pro-inflammatory conditions and explore the dynamics of nanodomain form by P2X7.
