Transition de Phase Liquide-Liquide, interactions en solution et cristallisation dans le cas du BPTI

Abstract : At the moment the achievement of good quality crystals is the bottleneck for the determination of protein structures. To solve this problem, searchers are studying the physical mechanisms of protein crystallization. Consequently, they study exhaustively “model” systems, among which the Bovine Pancreas Trypsin Inhibitor (BPTI). One of the properties of BPTI is the presence in the crystallization domain of a metastable Liquid-Liquid phase separation. The aim of this thesis is to understand the correlations between Liquid-Liquid and Liquid-Solid transitions. Therefore, we analyze by SAXS oligomerization and interactions between the different BPTI oligomers, as a function of physico-chemical parameters such as pH, temperature and the crystallizing agent. We propose an explanation of the crystallization properties of the BPTI.
