ETUDE DE LA DISSOLUTION ET DE LA CROISSANCE DE PRINCIPES ACTIFS

Abstract : In the pharmaceutical industry, dissolution is an important phenomenon that can determine the rate of adsorption of the active ingredient into the human body. Here we study, experimentally, the dissolution of two active ingredients before modelling the dissolution in solution by population balances. First, we developed a technique for continuous in situ monitoring of the dissolution of active ingredients using the conductivity technique. This allowed us to present a qualitative study of the phase transition of irbesartan where we highlight the different stages that characterize this transition as well as the influence of the habit and size distribution of the crystals used. Then we present the study of the dissolution of irbesartan A. By working on the initial dissolution rates we highlight two dissolution behaviours. For large underaturations, dissolution is controlled by material transfer while for small underaturations dissolution is controlled by surface phenomena. The influence of mass, media agitation and temperature on dissolution behaviour was also studied to guide and properly address the modelling part of the dissolution of irbesartan A. Finally, in the last part we model the dissolution of active ingredients by population balances. By monitoring the concentration and considering the fragmentation of the crystals, it was possible to satisfactorily model the dissolution.
