Quantitative biodistribution and Pharmacokinetics of theranostic multimodal gadolinium-based nanoparticles for lungs using Ultrashort-TE MRI

Résumé

Lung has always represented a challenge for MR imaging because of its intrinsic properties. In the last years, the development of new fast imaging sequences like the Utrashort Echo-Time (UTE) has opened new perspectives in the field[1], renewing the interest in MRI contrast media applied to the lung[2]. In this work we propose an UTE MRI study of the biodistribution and pharmacokinetics of two intratracheally administrated Gd-based contrast agents, namely the gadoteric acid (Dotarem) and ultra-small (<5nm) multimodal theranostic nanoparticles (USRPs)[3]. The pharmacokinetics, biodistribution and concentration values of the USRPs were validated against optical imaging of fluorescent USRPs. In relation to the pharmacokinetics of the contrast agents, an acute toxicological study was performed as well in order to evaluate the short-term toxicity of the USRPs.
