Caractérisation fonctionnelle des gènes homologues de CYCLOIDEA identifiés chez Aquilegia coerulea ‘Origami’.

Abstract : In Antirrhinum majus flower, zygomorphy (or floral bilateral symmetry) is determined by the interaction of 4 factors of transcription. The most important is coded by the CYCLOIDEA gene (CYC). CYC is expressed in the dorsal part of the flower, where it slows down the growth of organs. CYCLOIDEA homologs were characterized in 5 species of Ranunculaceae. Among these species, Aquilegia (which is actinomorphic) has 2 CYC homologous genes which are expressed in floral buds. The objectives of my work were to estimate the role of these genes in floral morphology and to prepare future complementary experiments. To do so, I used a method of gene silencing, the VIGS (Virus Induced Gene Silencing), that I applied to differents species of Ranunculaceae.The silencing of the CYC homologs did not modify the floral symmetry of Aquilegia but it seems to result in homeotics conversions (sepals to petals and stamens to petals). We deduced that, in Aquilegia, these genes might play a role in the regulation of the genes determining the floral organs identities or in determining the limits of their action area. In addition, we obtained promising results on the others tested species.
